Polilli was born in Osceola Mills, Pennsylvania, USA, on 28 March 1906, to parents Anthony Marino and Josephine Aveni. Both of her parents were born in Italy.
At some point, she married Louis A. Polilli (1903–1990). The couple had two children: Joe Polilli and Rosemary Natale.
Her husband passed away on 24 August 1990. She was a dedicated member of St. Joseph Catholic Church in Dover, where she actively participated in the LCBA and Christian Mothers organizations. She also devoted 25 years of her life to volunteering for the Community Hospice of Tuscarawas County.
Polilli passed away in Dover, Ohio, USA, on 4 February 2017, at the age of 110 years, 313 days. She was survived by her daughter, nine grandchildren, 21 great-grandchildren and 12 great-great-grandchildren.
At the time of death, she was the third-oldest (known) living person in the U.S. state of Ohio, behind Lessie Brown and Lela Schnorf.
